pharmachologic effect

The active substance of the drug reduces the ability of platelets to stick together, thereby improving blood circulation in small vessels. In addition, Trental reduces the protein content of fibrinogen, which significantly affects the viscosity of the blood and improves the state of the cellular membranes of red blood cells, increasing their resistance to deformation.

There is also a general relaxing effect of the drug on the vascular wall, which allows to reduce resistance and increase the amount of blood arriving to the organs. Also, the drug improves the functional state of the central nervous system. Trental has no effect on the heart rate.

The therapeutic effect is achieved an hour after taking the drug in the form of tablets. Its neutralization occurs in the liver, excretion through the kidneys. Decomposition products also have a curative effect and have a similar effect.

Indications for use

What are these tablets from? Trental has the following indications:

Contraindications

The use of tablets Trental 100 mg is contraindicated in certain pathological and physiological conditions of the patient's body, which include:

  1. Increased blood pressure (arterial hypertension), which is difficult to treat with drugs.
  2. Hypersensitivity to any chemical compounds of methylxanthine derivatives.
  3. Hypersensitivity to any of the components of the drug.
  4. Volumetric bleeding of various localizations in the body.
  5. Acute myocardial infarction (death of the heart muscle).
  6. Extensive hemorrhages localized in the retina of the eye.
  7. Severe violations of the rhythm of contractions of the heart (arrhythmia).

Dosage and route of administration

As indicated in the instructions for use, Trental is taken internally. Tablets should be swallowed whole, washed down with sufficient water, during or immediately after meals.

The average dose of the drug for oral administration is 100 mg (1 tab.) 3 times / day, followed by a slow increase in the dose to 200 mg (2 tablets) 2-3 times / day. The maximum single dose is 400 mg. The maximum daily dose is 1200 mg.

In patients with impaired renal function (QC less than 30 ml / min), the dose can be reduced to 100-200 mg (1-2 tablets) / day.

Dose reduction, taking into account individual tolerance, is necessary in patients with severe impairment of liver function.

Treatment can be initiated in small doses in patients with low blood pressure, as well as in patients at risk because of possible reduction of blood pressure (patients with severe forms of ischemic heart disease or with hemodynamically significant stenoses of cerebral vessels). In these cases, the dose can be increased only gradually.

Side effects

The use of high doses of Trental or an IV infusion exceeding the recommended infusion rate can cause the following side effects on the part of organs and systems:

  • sensory organs: scotoma, visual impairment;
  • nervous system: dizziness, headache, convulsions, sleep disorders, anxiety;
  • digestive system: anorexia, xerostomia, intestinal atony;
  • Cardiovascular system: arrhythmia, tachycardia, cardialgia, decreased blood pressure, progression of angina pectoris;
  • hypersensitivity reactions: skin hyperemia, itching, urticaria, anaphylactic shock, angioedema;
  • skin and subcutaneous fatty tissue: swelling, hyperemia of the skin of the face, increased fragility of the nails, tides of blood to the face and to the upper chest;
  • system of hemostasis and hematopoiesis: thrombocytopenia, leukopenia, pancytopenia, bleeding (from the mucous membranes, vessels of the skin, intestines, stomach), hypofibrinogenemia.

Very rarely there were cases of intrahepatic cholestasis, an increase in the activity of liver enzymes, and the emergence of aseptic meningitis.

Overdose

Trental can potentiate the effect of antihypertensive drugs.

In combination with ketorolac, the risk of bleeding and / or an increase in prothrombin time is increased; with meloxicam - increased risk of bleeding; with sympatholytic, ganglioblokatorami and vasodilators - it is possible to lower blood pressure; with heparin, fibrinolytic drugs - strengthening anticoagulant action.

Cimetidine increases the concentration of pentoxifylline in the blood plasma and increases the risk of side effects.
