Form of issue and composition

The form of release - tablets of 50 mg. 2 or 6 blisters of 10 tablets together with the instruction are placed in a separate cardboard package.

  • 1 tablet contains 50 mg of tofizopam, excipients are stearic acid, magnesium stearate, gelatin, talc, complex carbohydrates (lactose monohydrate, potato starch, microcrystalline cellulose).

Externally, Grandaxin is a round, flat disk-shaped tablets of white color, sometimes with a grayish hue, there is a chamfer, with a risk on one side tablets and the inscription "GRANDAKH" on the other hand, almost odorless.

Pharmacological effect

Grandaxin is a tranquilizer of the benzodiazepine series. The main action is anxiolytic (soothing). Unlike other benzodiazepine tranquilizers, it does not have sedative, hypnotic, miorelaxing, anticonvulsant effects.

The medication regulates psychovegetatics, relieves of various vegetative disorders. Has a moderate stimulating effect. These pharmacological properties cause the drug to belong to daytime anxiolytics. Due to the absence of a muscle-relaxing effect, the drug can be taken with patients with myasthenia gravis and myopathy.

Atypicality of the chemical formula Tofizopama, which is very different from the structure of the classic anxiolytic benzodiazepine series, gives the drug a number of advantages:

  • in the therapeutic dose does not enhance the effect of ethanol;
  • does not cause withdrawal syndrome;
  • does not cause mental or physiological addiction.

The mechanism of action is associated with a stimulating effect on benzodiazepine receptors in the brain.   6. Neuroses and pseudo-neurotic conditions (neuropsychiatric disorders, characterized by emotional stress, moderate increased anxiety, apathy, decreased activity, the appearance of compulsive experiences, autonomic disorders);
  7. Neurogenic muscular atrophy, myopathy, myasthenia gravis and other pathological conditions that are accompanied by secondary neurotic symptoms and in which the use of anxiolytics (tranquilizers), which have a pronounced myorelaxing effect, is contraindicated;
  8. Pathological menopause (in the annotation it is indicated that with a menopausal syndrome the drug can be prescribed both as an independent agent and as an adjunct to hormonal therapy).

Contraindications

Contraindications to taking medication are:

  1. Decompensated respiratory failure.
  2. Period of pregnancy (in particular, I trimester) and breastfeeding (lactation).
  3. Hypersensitivity to the active ingredient or other auxiliary components of the drug.
  4. Conditions accompanied by aggressiveness, irritability, marked psychomotor agitation or depression.
  5. Joint use with cyclosporine, sirolimus, tacrolimus.
  6. Congenital lack of lactase, galactose intolerance or malabsorption syndrome of galactose and glucose (Granaxin includes lactose monohydrate).
  7. Syndrome of stopping breathing in a dream (available in a patient's history).

Grandaxin is administered with extreme caution in: acute respiratory failure in a patient's history, epilepsy, angle-closure glaucoma, decompensated chronic respiratory distress, organic brain damage (for example, with atherosclerosis).

Side effects

Overdose

When ingestion of Grandaxin in high doses (50-120 mg / kg body weight) symptoms of oppression of functions can be observed CNS: blurred consciousness, vomiting, oppression of the respiratory center and / or epileptic seizures, coma.

If there is severe suppression of the CNS functions, do not induce vomiting. Gastric lavage is recommended. The purpose of activated carbon can reduce absorption of the drug. Introduction of CNS stimulants is not recommended.

Hypotension is preferably eliminated by intravenous administration of fluids and by placing the patient in the Trendelenburg position. If these measures do not lead to normalization of blood pressure, the administration of norepinephrine (noradrenaline) or dopamine is allowed. The effectiveness of dialysis and forced diuresis is not proven. In extreme cases, it is possible to use flumazenil, a specific antagonist of benzodiazepine, but its use in an overdose of Tofizopam has not been adequately studied.

  2. Each tablet of Grandaxin contains 92 mg of lactose, which should be taken into account for patients with lactose intolerance.
  3. In patients with epilepsy, tofisopam may increase the threshold of convulsive readiness.
